چکیده

the pharmacokinetics of recombinant human erythropoietin (rhuepo) administered intravenously has been investigated in a group of five patients with chronic renal failure who were dialysis-dependent. the half-life of circulating erythropoietin decreased from 7.9±oa hr (mean ±so) at the beginning of treatment to 6.2±o.6 hr after 6 weeks and 5a±o.9 hr after 4 months of treatment. in spite of the sustained increase in haemoglobin neither the red ce1l2,3,-diphosphoglycerate (2,3- dpo) nor the p50 decreased from the pretreatment values. the mechanism which governs the rise in 2,3-diphosphoglycerate in chronic renal failure is different from the adaptive mechanism which operates in other types of anaemia.

PHARMA COKINETICS OF RECOMBINANT ERYTHROPOIETIN AND RED CELL METABOLISM IN HAEMODIALYSIS PATIENTS

In spite of t...

Resistance to recombinant human erythropoietin therapy in haemodialysis patients

The involvement of a humoral factor (named as haemopoietin) in the regulation of haematopoiesis, was firstly described in literature in 1906 (Carnot  Deflandre, 1906). However, only 40 years later a linkage between erythropoietin (EPO) and erythropoiesis was described (Bondsdorff  Jalavisto, 1948), and only in the 1950s was established that the kidney is the main site of production of EPO (Ja...

Long-term therapy with recombinant human erythropoietin increases CD8+ T-cell apoptosis in haemodialysis patients.

BACKGROUND We intended to assess the intensity of apoptosis in the CD4+ and CD8+ T-lymphocytes of haemodialysis (HD) patients on recombinant human erythropoietin (rHuEpo). METHODS The expression of Fas, tumour necrosis factor-alpha receptors (TNFRI and TNFRII) and the CD28 molecule on lymphocytes was evaluated in 15 HD patients before and during treatment with rHuEpo. In cultures of periphera...

Benefits and risks of protracted treatment with human recombinant erythropoietin in patients having haemodialysis.

Fourteen patients with uraemic anaemia and having regular haemodialysis were given human recombinant erythropoietin in increasing doses, beginning with 24 U/kg thrice weekly. One patient was dropped from the study because of recurrent thrombosis of vascular access sites. In the other 13 patients, followed up for a mean of 9.1 months (range 8-11), haemoglobin concentrations increased from 62 (SD...

Hepcidin serum levels and resistance to recombinant human erythropoietin therapy in haemodialysis patients.

OBJECTIVE The aim of this study was to analyze the factors that are associated with the response to erythropoiesis-stimulating agents (ESAs) and its association with hospitalization and mortality rates; to evaluate the serum hepcidin level and its associations with iron profile, inflammatory markers, ESA responsiveness, and mortality; and to determine independent factors affecting ERI and hepci...

The Effect of Erythropoietin on Rat’s Red Blood Cell Indices in Simulated Microgravity (Experimental Study)

Purpose: Microgravity causes major changes in various systems of the body in space, such as shift in cerebrospinal fluid, decreased red blood cells count, and electrolyte loss. These complications are very important in space and we should find new approaches to prevent the side effects of microgravity in astronauts. Materials and Methods: This experimental study was conducted on 21 adult ma...
